| Description | Neuronal growth regulator-1 (NEGR-1) is a 46 kDa member of immunoglobulin superfamily of cell adhesion molecules (IgLONs). IgLONs are the most abundant glycoylphosphatidylinositol-anchored proteins expressed in neurons and they include limbic system-associated membrane molecule (LSAMP), neurotrimin (NTM), opioid-binding cell adhesion molecule (OBCAM), and NEGR-1. IgLON proteins contain three immunoglobulin domains, GPI anchor, and 6-7 potential glycosylation sites. IgLONs form hemophilic and heterophilic complexes to modulate cell adhesion and neurite outgrowth. IgLONs play a role in the formation and maintenance of excitatory synapses. NEGR-1 is expressed in the brain, specifically in the cerebrum, brain stem, hippocampus, and cerebellum. It may also function as a trans-neural growth-promoting factor in regenerative axon sprouting in the mammalian brain. Genome-wide analysis revealed that NEGR1 is associated with human body weight regulation. NEGR1 is also frequently downregulated in various human cancers and is a substrate for metalloproteinase-dependent shedding. |
