| Description | Filaggrins are an important class of the intermediate filament-associated proteins which interect with keratin intermediate filaments (Ifs) of terminally differentiating mammalian epidermis. Filaggrins exhibit wide species variations. Filaggrin is expressed only in well-differentiated keratinized epithelial cells. The synthesis of filaggrin varies in types of ichthyosis, and with viral, premalignant and malignant conditions. Filaggrin also appears to be a target molecule for rheumatoid arthritis-specific auto-antibodies in humans. Keratohyalin granules in the granular layer of interfollicular epidermis are predominantly composed of the 400-kD protein profilaggrin. Following a short, unique N-terminal domain, most of the profilaggrin molecule consists of 10 to 12 repeats of the 324-residue filaggrin sequence. Upon terminal differentiation of granular cells, profilaggrin is proteolytically cleaved into filaggrin peptides of approximately 37 kD and the N-terminal domain containing an S100-like calcium-binding domain.Synonyms: FLG |
