Directions for use |
- Take 100 µl peripheral blood anticoagulated by EDTA and add to the bottom of a 5 ml tube.
- Add 10 µl labeled antibody to the bottom of the flow tube and mix with the whole blood. Allow to stand for 20 minutes at room temperature in the dark.
- Add 2 ml RBC lysis buffer, mix, then incubate for 10 minutes in the dark to dissolve red blood cells.
- Centrifuge the sample tube at 1000 RPM for 5 minutes. Discard the supernatant.
- Add 2 ml PBS to resuspend and wash the cells, then centrifuge at 1000 RPM centrifugation for 5 minutes. Discard the supernatant.
- Add 0.5 ml PBS to resuspend the cells, and detect by flow cytometry (sample should be measured on the same day, or can be fixed overnight at 4 °C then measured).
