Description | Intercellular Adhesion Molecule-3 (ICAM-3, CD50) is a highly glycosylated protein that belongs to immunoglobulin superfamily. It has five extracellular Ig-like domains and binds the leukocyte integrins LFA-1 (CD11a/CD18) and αdβ2. ICAM-3 is broadly expressed on leukocytes, endothelial cells, and Langerhans cells. ICAM-3 is an adhesion molecule with signal transducing functions involved in immune responses. LFA-1/ICAM-3 interactions between APC and resting T cells provide costimulatory signals that are important for initiating immune responses. LFA-1/ICAM-3 interactions can also exert inhibitory effects on proliferating T cells. ICAM-3 induces intracellular Ca2+ mobilization and undergoes phosphorylation on tyrosine residue leading to association with the tyrosine phosphatase, CD45. Soluble forms of ICAM-3 have been found to be elevated in the circulation of patients with autoimmune disorders. Alternative splicing results in multiple transcript variants encoding different isoforms. Diseases associated with ICAM3 include Actinic Prurigo and Trichosporonosis. |
