Description | IGF-BP7 belongs to the insulin-like growth factor-binding protein family. It shares 20 - 25% identity with other members of the family. IGF-BP7 contains the common IGFBP motif (GCGCCXXC) at the N-terminal, in a region containing the cysteine-rich domain that is conserved among the IGFBPs. It also possesses a C-terminal Ig-like domain. IGFBPs bind IGF peptides with high affinity and regulate the biological activities of IGFs. IGF-BP7 binds to IGF-I, IGF-II, and insulin (with very low affinity). IGF-BP7 also binds to IGF-1R and blocks its activation by insulin-like growth factors as well. Results from extended incubations with IGF-BP7 indicate that there is an activation of the translational repressor 4E-binding protein 1 (4E-BP1), enhanced sensitivity to apoptosis in IGF1R-positive cells, and subsequent inhibition to protein synthesis, cell growth, and survival in the treated cells. IGF-BP7 is abundantly expressed by normal tissues; nevertheless, no or very low expression is detected in advanced stages of many cancers. The expression of IGF-BP7 correlates with favorable prognosis in colon cancer patients. IGF-BP7 may have a potential tumor suppressor role in the treatment of different types of cancer, including cervical cancer, osteosarcoma, prostate cancer, breast cancer, lung cancer, melanoma, thyroid cancer, and hepatocellular carcinomas. |
