Description | DLL1 (Delta-like protein 1) is a type I membrane protein that belongs to the DSL (Delta/Serrate/Lag2) family of Notch ligands. It is required for embryonic development and maintenance of adult stem cells. There are five Notch ligands (DLL1, DLL3, DLL4, Jagged-1 and Jagged-2) and four Notch receptors (Notch-1 to Notch-4) in mammals. DLL1 is widely expressed and mouse DLL1 shares 91% and 95% amino acid sequence identity with human and rat DLL1 respectively. Interactions of Notch receptors with their ligands result in sequential proteolysis by ADAM (a disintegrin and metalloprotease) and then by presenilin/gamma secretase, resulting in the shedding of the extracellular domains and the generation of soluble ICD (intracellular domain) signaling fragments which translocate to the nucleus to interact with transcriptional factors. DLL1 undergoes proteolytic processing in its extracellular domain by ADAM10 in a similar manner to the Notch receptor, and its ICD may participate in bidirectional signaling. DLL1-induced Notch signaling regulates cell lineage, cell specification, cell patterning and morphogenesis through its effects on differentiation and proliferation. DLL1 plays an important roles in brain development at many different levels. During cerebellar development, DLL1 is required for the formation of the Bergmann glial layer and its morphological maturation. During neocortex development, DLL1-Notch signaling coordinates progenitor cell division and differentiation across radial and zonal boundaries. |
