Description | TNFRSF12A was initially identified as a fibroblast growth factor-1-inducible gene in murine NIH 3T3 cells, and was named as fibroblast growth factor-inducible-14 (Fn14) gene. Human TNFRSF12A was cloned from a HUVEC cDNA library and identified as the TWEAK receptor and a member of the TNFR family. It is a type-I transmembrane protein of 128 amino acids in length and has a cysteine rich region in the extracellular domain and a TRAF-binding motif in the intracellular domain. TNFRSF12A binds to TRAF2 (TRAFs 1, 3, and 5) in yeast two-hybrid screen and can initiate both canonical and noncanonical NF-κB activation. Also, TNFRSF12A-TNFRSF12 signaling induces lysosomal degradation of a cIAP1–TRAF2 complex to sensitize tumor cells to TNFRSF12A and its ligand trigger proliferation of mesenchymal progenitor cells and inhibit differentiation of chondrocytes, osteoblasts, and myocytes. TNFRSF12A expression is upregulated on epithelial and mesenchymal progenitor cells after tissue damage induced by including hypoxia, oxidative stress, chemical and mechanical injuries, inflammation, and tumor growth. TNFRSF12A and its ligand play a key role in muscle atrophy, cerebral ischemia, kidney injury, atherosclerosis, experimental autoimmune encephalitis, rheumatoid arthritis, and inflammatory bowel disease. |
