| Description | MMP-3 ELISA Assay Kit is based on the simultaneous binding of human MMP-3 by two different monoclonal antibodies, one coated on the microplate and the other conjugated with biotin; after the incubation, the unbound reagent is washed away through a washing step. Then the Streptavidin conjugated with the horseradish peroxidase (HRP) is added; it recognizes and bounds to the biotin; after the incubation, the unbound reagent is washed away through a washing step. Following these incubations, a complex between the Antigens, the biotin, the streptavidin and the peroxidase is formed. The peroxidase in the bound-fraction reacts with the Substrate (H2O2) and the TMB Substrate and develops a blue color that changes into yellow when the Stop Solution (H2SO4) is added. The color intensity is proportional to the MMP-3 concentration in the sample. MMP-3 concentration in the sample is calculated through a calibration curve. Sample Types: serum, plasma. Range: 31.25 - 2000 ng/ml. Sensitivity: 4 ng/ml. Time: 2 hours • Rheumatoid Arthritis (RA) is one of the most common autoimmune diseases (1-2% of European population). A major focus of medical research is to identify this pathology in its early stages to develop a targeted drug therapy since its initial stages. In this sense, one of the most useful markers is the MMP-3 (Matrix Metalloproteinase 3), because it allows both the identification of RA in its early stages, both the monitoring of the patient conditions during the drug therapy: in fact, during the development of RA, serum levels of MMP-3 are raised with respect to the physiological state of the healthy subject, and tend to decrease during the drug treatment of the patient. |
