Description | Uteroglobin, also known as Clara Cell-specific 10-KD protein (CC10) is the first member of Secretoglobin superfamily of proteins. Uteroglobin is a small globular, non-glycosylated, secreted protein that was first discovered in the rabbit uterus. Uteroglobin is produced by mucosal epithelial cells in the lung (Clara cells), uterus and prostate. It is a homodimer formed by two 70 amino acid subunits joining in antiparallel orientation by disulfide linkage. This homodimer forms a large central hydrophobic cavity that binds hydrophobic ligands such as phosphatidylcholine, phosphatidylinositol, polychlorinated biphenyls (PCB) and progesterone. Sequestering of carcinogens such as PCB is anti-tumorigenic. Uteroglobin is a steroid-inducible protein with many biological activities including potent anti-inflammatory activity mediated through the inhibition of soluble phospholipase A2 activity. Uteroglobin inhibits fMLP induced chemotaxis of monocytes and neutrophils, and also inhibits the recognition of embryonic and sperm antigens by lymphocytes. Uteroglobin binds with high affinity to specific receptor sites on several cell types, and inhibits cellular invasion of the extracellular matrix. Uteroglobin is suggested to be a protective mediator in the airway inflammatory process and to have protective effects on the respiratory tract from oxidative stress. A single nucleotide polymorphism in the UG gene is associated with several inflammatory/autoimmune diseases. |
