Description | Matrix metalloproteinases (MMPs) are a family of zinc-dependent endopeptidases that degrade components of the extracellular matrix (ECM) and play essential roles in various physiological and pathological processes such as morphogenesis, differentiation, angiogenesis, tissue remodeling, and tumor invasion. A typical MMP consists of a pro-peptide of about 80 amino acids, a catalytic metalloproteinase domain of about 170 amino acids, a linker peptide of variable lengths and a hemopexin domain of about 200 amino acids. The zinc binding motif HEXXHXXGXXH in the catalytic domain, and the ‘‘cysteine switch’’ motif PRCGXPD in the pro-peptide are common structural signatures. Matrix metalloproteinase 10 (MMP-10, stromelysin 2) degrades a broad range of substrates including gelatin, collagen types III, IV and V, fibronectin, aggrecan, and pig cartilage proteoglycan. MMP-10 can activate other MMPs such as MMP-1 and MMP-8. It is expressed by macrophages in numerous tissues after injury. MMP-10 expressed in macrophages plays as a facilitator of cell migration and invasion. Its expression from macrophage may have beneficial effect in response to acute infection based on the mouse model study of human lung cystic fibrosis. The expression of MMP-10 from macrophages in this model increases survival of mice with minimal weight loss after Pseudomonas aeruginosa acute infection. In lung cancer, MMP-10 is required for maintenance of a highly tumorigenic, cancer-initiating, metastatic stem-like cell population. It also plays a role in the maintenance of cancer stem-like cells and cancer-initiating cells of epithelial ovarian cancer by activating canonical Wnt signaling through inhibition of noncanonical Wnt signaling ligand Wnt5a. In muscle, MMP-10 participates in proper repairing after ischemia. |
